Murphy will be matching Martin Corry's mark against the Welford Road visitors from Northern Ireland.
Full-back Murphy leads a team that sees Horacio Agulla reclaim the number 14 shirt, while England scrum-half Ben Youngs has regained full fitness following the World Cup and is on bench duty.
Tigers won their Pool Four opener away to Aironi last weekend, although it was achieved without the bonus point most pundits had expected.
"We should have got more out of the game," said Murphy. "But full credit to Aironi, they defended well and were aggressive.
"There is a long way to go, and we might have to pick up bonus points later in the season, but hopefully it won't come back to haunt us."
Ulster show two changes from the side that accounted for Clermont Auvergne in Belfast last Saturday, with Nevin Spence replacing injured centre Paddy Wallace and Declan Fitzpatrick taking over from prop John Afoa, who has made a flying return to New Zealand for best man duties at All Blacks colleague Jerome Kaino's wedding.
Ulster's starting XV includes former Leicester fly-half Ian Humphreys, who scored all his team's points in the stirring 16-11 success against Clermont.
"We will be travelling to Welford Road full of confidence and thinking we can win that game," Humphreys said.
"We are not thinking 'we will go and give it a crack, we might get close and maybe get a losing bonus point,' we are now going over there thinking there is no reason why we cannot get the four points."
