England's 2015 World Cup organising committee have announced Deighton and former England captain Bill Beaumont as its latest board member additions.
It means that Deighton will link up again with former LOCOG director of sport Debbie Jevans, now England Rugby 2015's chief executive.
Deighton's Olympics role included leading LOCOG's day-to-day operations and having responsibility for a £2billion budget from the private sector.
"Everyone recognises that the London Olympics and Paralympics have set a high benchmark for international sports events that follow," Deighton said.
"I want to bring the knowledge and experience gleaned from my time with LOCOG to help ensure that the organisation, delivery and feelgood factor of the Rugby World Cup in 2015 is right up there with London 2012."
Beaumont, the current Rugby Football Union and Six Nations chairman, is a former Rugby World Cup director and current International Rugby Board council member.
Deighton and Beaumont will join a board that currently comprises Jevans, chairman Andy Cosslett, Ian Metcalfe, Ian Watmore, Ian Ritchie, Stephen Brown, Geoff Irvine and Quentin Smith.
"Our job at England Rugby 2015 is to build the strength and appeal of this great game by delivering a brilliant Rugby World Cup," Cosslett said.
"To do this we need to call on the talents and experience of people drawn from both inside and outside the world of rugby.
"I am delighted to welcome Bill and Paul to the board."
