Van Wyk, who came on as a first-half replacement in the heartbreaking 22-23 loss to the Sharks last week, will start ahead of Ricardo Croy - despite the latter recovering from injury.
With Van Wyk in the No.10 jersey, Tiaan Marx moves to inside centre in place of injured SA Sevens star Pieter Engelbrecht.
Loosehead prop Dawie Steyn has recovered from a leg injury and returns to the starting XV, in place of De-Jay Terblanche - who drops down to the bench.
